== Early life and education == Gavin Christopher Newsom was born on October 10, 1967, in San Francisco, California, to Tessa Thomas (née Menzies) and William Alfred Newsom III, a state appeals court judge and attorney for Getty Oil. He is a descendant of the Newsom family of architects active in early 20th century California, who originally came from Canada. William Newsom was a close friend of billionaire heir Gordon Getty, who repeatedly supported Gavin's career. William Newsom would also for a time help manage the Getty Trust. A fourth-generation San Franciscan, Newsom comes from a prominent family. His maternal great-grandfather Thomas Addis was a nephrologist and professor of medicine at Stanford University noted for his research on kidney disease. Newsom is a second cousin twice removed of musician Joanna Newsom, and his aunt Barbara Newsom was married to Ron Pelosi, the brother-in-law of former House speaker Nancy Pelosi. Newsom's parents divorced in 1971 when he was three years old, leaving his mother, Tessa, to raise him and his younger sister, Hilary Newsom Callan, largely on her own. Tessa worked three jobs—often as a waitress, bookkeeper, and secretary—to support the family, and Newsom has cited her as shaping his work ethic. Newsom has called his childhood challenging, shaped in part by financial instability and his struggle with "pretty severe" dyslexia, a condition he still has. === Identifying Translated mRNA Regions === By using specific drugs, ribosome profiling can identify initiating regions of mRNA, elongating regions, and areas of translation stalling. Initiating regions can be detected by adding harringtonine or lactidomycin to prevent any further initiation. This allows the starting codon of the mRNAs throughout the cell lysate to be analyzed, which has been used to determine non-AUG sequences that do initiate translation. The other elongating regions can be detected by adding antibiotics like cycloheximide that inhibit translocation, chloramphenicol that inhibits transfer of peptides within the ribosome, or non-drug means like thermal freezing. These elongation freezing methods allow for the kinetics of translation to be analyzed. Since multiple ribosomes can translate a single mRNA molecule to speed up the translation process, RiboSeq demonstrates the protein coding regions within the mRNA and how quickly this is done depending on the mRNA being sequenced. This also allows for ribosome profiling to show pause sites within the transcriptome at specific codons. These sites of slow or paused translation are demonstrated by an increase in ribosome density and these pauses can link specific proteins with their roles within the cell.

=== Pharmacokinetics === Gepotidacin is rapidly absorbed orally, reaching peak plasma concentrations (tmax) after approximately 2.0 hours. In adults with uncomplicated urinary tract infections (uUTI) and normal renal function, the mean steady-state maximum concentration (Cmax) is 4.2 mcg/mL, and the area under the concentration-time curve over 12 hours AUC(0-12) is 22.8 mcg*hour/mL following a 1500 mg dose every 12 hours. Systemic exposure (Cmax and AUC) increases proportionally with dose. Accumulation of approximately 40% occurs and achieves a steady state by day 3. The absolute bioavailability is about 45%, and standard and moderate fat meals did not significantly affect its absorption. Gepotidacin's pharmacokinetics were found to be generally consistent across different ages, sexes, races, and body weights during modeling and simulation. Gepotidacin has a mean steady-state volume of distribution (Vss) of 172.9 liters and is 25-41% bound to plasma proteins. It has a terminal elimination half-life of approximately 9.3 hours and a total clearance of 33.4 L/hour. The primary metabolic pathway involves CYP3A4, with a minor metabolite (M4, ~11% of circulating drug). The co-administration of other drugs can influence gepotidacin levels. Strong inhibitors of CYP3A4 can increase gepotidacin exposure, whereas strong inducers of CYP3A4 can decrease it. ==== Excitation-contraction coupling ==== Excitation contraction coupling is the process by which a muscular action potential in the muscle fiber causes the myofibrils to contract. This process relies on a direct coupling between the sarcoplasmic reticulum calcium release channel RYR1 (ryanodine receptor 1), and voltage-gated L-type calcium channels (identified as dihydropyridine receptors, DHPRs). DHPRs are located on the sarcolemma (which includes the surface sarcolemma and the transverse tubules), while the RyRs reside across the SR membrane. The close apposition of a transverse tubule and two SR regions containing RyRs is described as a triad and is predominantly where excitation–contraction coupling takes place. Excitation–contraction coupling occurs when depolarization of skeletal muscle cell results in a muscle action potential, which spreads across the cell surface and into the muscle fiber's network of T-tubules, thereby depolarizing the inner portion of the muscle fiber. Depolarization of the inner portions activates dihydropyridine receptors in the terminal cisternae, which are close to ryanodine receptors in the adjacent sarcoplasmic reticulum. The activated dihydropyridine receptors physically interact with ryanodine receptors to activate them via foot processes (involving conformational changes that allosterically activates the ryanodine receptors). As the ryanodine receptors open, Ca2+ is released from the sarcoplasmic reticulum into the local junctional space and diffuses into the bulk cytoplasm to cause a calcium spark.
=== Lipid separation === The simplest method of lipid separation is the use of thin layer chromatography (TLC). Although not as sensitive as other methods of lipid detection, it offers a rapid and comprehensive screening tool prior to more sensitive and sophisticated techniques. Solid-phase extraction (SPE) chromatography is useful for rapid, preparative separation of crude lipid mixtures into different lipid classes. This involves the use of prepacked columns containing silica or other stationary phases to separate glycerophospholipids, fatty acids, cholesteryl esters, glycerolipids, and sterols from crude lipid mixtures. High-performance liquid chromatography (HPLC or LC) is extensively used in lipidomic analysis to separate lipids prior to mass analysis. Separation can be achieved by either normal-phase (NP) HPLC or reverse-phase (RP) HPLC. For example, NP-HPLC effectively separates glycerophospholipids on the basis of headgroup polarity, whereas RP-HPLC effectively separates fatty acids such as eicosanoids on the basis of chain length, degree of unsaturation and substitution. For global, untargeted lipidomic studies it is common to use both RP and NP or Hydrophilic Interaction Liquid Chromatrography (HILC) columns for increased lipidome coverage. The application of nano-flow liquid chromatography (nLC) proved thereby to be most efficient to enhance both general measurement sensitivity and lipidome coverage for a global lipidomics approach.

In an incomplete elution, part of the analytes remain in the sorbent because the eluent used does not have a strong enough affinity. Many of the adsorbents/materials are the same as in chromatographic methods, but SPE is distinctive, with aims separate from chromatography, and so has a unique niche in modern chemical science.

There will be no "ideal" site. The "best possible" site will be sought. Mining areas and regions where volcanoes have been active or where there is a risk of earthquakes are excluded. Internationally, experts are advocating storage in rock formations several hundred meters below the earth's surface. This involves building a repository mine and storing the waste there. It is then permanently sealed. Geological and technical barriers surrounding the waste are designed to keep it safe for thousands of years. For example, 300 meters of rock will separate the repository from the earth's surface. It will be surrounded by a 100-meter-thick layer of granite, salt or clay. The first waste is not expected to be stored until 2050. The Federal Office for the Safety of Nuclear Waste Management (BfE) took up its activities on September 1, 2004. Its remit includes tasks relating to nuclear safety, the safety of nuclear waste management, the site selection procedure including research activities in these areas and, later on, further tasks in the area of licensing and supervision of repositories.
